ABSTRACT

The cleft lip and palate (CLP) is one of the most common craniofacial malformations in humans. We collected functional magnetic resonance data of 23 CLP patients before rehabilitation training (Bclp) and 23 CLP patients after rehabilitation training (Aclp), who were performing Chinese character pronunciation tasks, and performed brain activation analysis to explore the changes of brain mechanism in CLP patients after articulation disorder rehabilitation training. The study found that Aclp group had significant activation in the motor cortex, Broca area, Wernicke area and cerebellum. While the Bclp group had weak activation in the motor cortex with a small activation range. By comparing the differences and co-activated brain regions between the two groups, we found that rehabilitation training increased the activity level of negatively activated brain areas (cerebellum, left motor area, Wernicke area, etc.) to a positive level. At the same time, the activity level of weakly activated brain areas (right motor area, Broca area, etc.) was also increased. Rehabilitation training promoted the activity level of articulation-related brain regions. So that the activation intensity of articulation-related brain regions can be used as a quantifiable objective evaluation index to evaluate the effect of rehabilitation training, which is of great significance for the formulation of rehabilitation training programs.

ABSTRACT

OBJECTIVE To optimize the existing t hin layer chromatography (TLC)identification and content determination methods of Jizhi syrup. METHODS High performance thin-layer chromatography (HPTLC)was used to identify five medicinal materials in Jizhi syrup ,such as Ilex chinensis ,Houttuynia cordata ,Peucedanum praeruptorum ,Citrus aurantium ,Glycyrrhiza uralensis. High performance liquid chromatography (HPLC)was used to determine the contents of procatechuic acid ,ephedrine hydrochloride and naringin in Jizhi syrup. RESULTS HPTLC results showed that the identification spots of pedunculoside , praeruptorin A ,naringin,and liquiritin were clearly displayed ,and the retention factors were in the range of 0.2 to 0.8. After validation,the method had been proved to be strongly specific ,robust and repeatable. HPLC results showed that the linear ranges of protocatechuic acid ,ephedrine hydrochloride and naringin were 4.32-431.67,1.14-114.17 and 7.02-702.33 μg/mL(all r> 0.996),respectively. The average recoveries were 100.61%,100.40% and 99.22%,and RSDs were all less than 2.00%. RSDs of precision(n=6),stability(24 h,n=7)and repeatability (n=6)were all less than 2.00%. The average contents of the three components in 10 batches were 623.3,152.1,1 213.9 μg/mL(RSD<10.00%),respectively. CONCLUSIONS In this study , HPTLC method of one-plate multi-drug is established for the identification of Jizhi syrup. One sample pretreatment method and two TLC conditions are used to realize the rapid identification of five kinds of medicinal materials. An HPLC method is established to determine the content of Jizhi syrup ,which realizes the fast quantification of three active components in Jizhi syrup ,and can be used to optimize the identification and content determination items in the existing legal quality standards of Jizhi syrup.

ABSTRACT

Objective: To investigate the analgesic substances in the aerial part of Urtica fissa (Urticae Fissae Herba), commonly used for rheumatoid and rheumatism arthritis. Methods: The analgesic constituents were isolated with the active guidance of hot plate and acetic acid writhing models, and identified by comprehensive spectroscopic analysis. Results: Thirteen alkaloids (1–13), two lignans (14, 15), and three amides (16–18) were isolated from the active fractions. Among them, compound 1 was a new alkaloid, and compound 6 was a new natural product. The activity evaluation in vivo indicated that various pyrrole alkaloids (1, 3, 6, and 12) possessed significant analgesic activities, they could significantly inhibit the mice pain response induced by acetic acid and hot plate at the dosage of 2 mg/kg BW. Conclusion: The study revealed that the pyrrole alkaloids played important roles in the analgesic activities of Urticae Fissae Herba.

ABSTRACT

To achieve uniform soluble expression of multiple proteins in the same Escherichia coli strain, and simplify the process steps of antigen production in genetic engineering subunit multivalent vaccine, we co-expressed three avian virus proteins including the fowl adenovirus serotype 4 (FAdV-4) Fiber-2 protein, infectious bursal disease virus (IBDV) VP2 protein and egg-drop syndrome virus (EDSV) Fiber protein in E. coli BL21(DE3) cells after optimization of gene codon, promoter, and tandem expression order. The purified proteins were analyzed by Western blotting and agar gel precipitation (AGP). The content of the three proteins were well-proportioned after co-expression and the purity of the purified proteins were more than 80%. Western blotting analysis and AGP experiment results show that all the three co-expression proteins had immunoreactivity and antigenicity. It is the first time to achieve the three different avian virus antigens co-expression and co-purification, which simplified the process of antigen production and laid a foundation for the development of genetic engineering subunit multivalent vaccine.

ABSTRACT

Objective To investigate the effect of low oxygen tension (7.5% O2) on human embryo development in vitro and outcome of women undergoing IVF-ET. Methods The data of 280 routine IVF cycles with fresh embryo transfer on day3 conducted in the 2nd People's Hospital of Yunnan Province from February 2012 to August 2013 were retrospectively analyzed. Two groups, 7.5%O2 low oxygen tension (n=120) and 20%O2 high oxygen tension (n=160),were setup for early embryo (D1-D3) development in vitro. Results Good quality embryo on day 3 in low oxygen tension group showed morphologically appearance of blastomere mutually clinging to each other,a characteristic of slight compaction. Higher rate of good quality embryo (66.22%V. 47.54%) and of clinical pregnancy (53.33%v. 39.22%) were found in the low oxygen tension group. There were no significantly differences in rates of fertilization,cleavage and embryo utilization respectively between the two groups ( all>0. 05) . Conclusion High quality early stage human embryos can be developed in vitro in low oxygen tension, and may improve clinic outcome of IVF.

ABSTRACT

<p><b>OBJECTIVE</b>To establish a RP-HPLC method for simultaneous determination of thymine, hypoxanthine and uracil contents in medical pipefish.</p><p><b>METHOD</b>Samples were extracted with distilled water by ultrasonic wave and separated on Waters C18 column eluted with a mobile phase of 0.05 mol x L(-1) KFI2PO4-acetonitrile (97:3). The flow rate was 0.6 mL x min(-1). The determination wavelength was 260 nm and the column temperature was set at 40 degrees C.</p><p><b>RESULT</b>The method had good linearity in the range of 0.033-0.660 (r = 0.9996), 0.620-12.400 (r = 0.9999), 0.048-0.960 microg (r = 0.9995), with average recoveries of 98.67% (RSD 1.6%), 99.03% (RSD 0.74%), 98.65% (RSD 1.8%), for thymine, hypoxanthine and uracil respectively.</p><p><b>CONCLUSION</b>The simultaneous determination method of thymine, hypoxanthine and uracil in medical pipefish is established by RP-HPLC for the first time. The contents of the three constituents in different kinds of medical pipefish are significantly different. The method is simple, rapid and sensitive, and can be used for control the quality of medical pipefish.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the identification method of Syngnathus and its adulterants.</p><p><b>METHOD</b>Random amplified polymorphic DNA (RAPD) was used to construct a dendrogram by UPGMA method based on Nei & Li's coefficient and a genetic affinity pattern for Syngnathus acus, Solenognathus hardwickii, Syngnathoides biaculeatus, Trachyrhamphus serratus, Halicampus koilomatodon, Microphis boaja.</p><p><b>RESULT</b>Four primers, LJ04, LJ09, LJ16 and LJ19, from 18 random primers were used in the dendrogram which can differentiate Syngnathus in genus level and showed a great consistence with the appearance identification. The genetic affinity pattern based on primers LJ09 and LJ19 could be used to identify Syngnathus from its adulterants.</p><p><b>CONCLUSION</b>RAPD is suitable to identify Syngnathus and its adulterants.</p>

ABSTRACT

Objective To study the chemical constituents of the EtOAC soluble fraction from Urtica fissa with obviously anti-inflammatory and analygestic effect. Methods The compounds were isolated by polyamide and silical gel colum chromatography and identified by UV, IR, NMR, and MS. Results Twelve compounds were isolated from U. fissa and identined as quercetin (Ⅰ), kaempferol (Ⅱ), kaempferol-3-O-?-D-glucopyranoside (Ⅲ), quercetin-3-O-?-D-glucopyranoside (Ⅳ), caffeic acid (Ⅴ), chlorogenic acid (Ⅵ), scopolin (Ⅶ), scopoletin (Ⅷ), kaempferol-3-O-rutinoside (Ⅸ), quercetin-7-O-?-D-glucopyranoside (Ⅹ), isorhamnetin-3-O-?-D-rutinoside (Ⅺ), ?-sitosterol ( ⅩⅡ ). Conclusion Compounds Ⅰ-Ⅺ are all isolated from U. fissa for the first time and compounds Ⅶ, Ⅹ, and Ⅺ are found in plants of Urtica L. for the first time also.
